Compound monograph — Sleep Mix

Sleep Mix is a five-component research blend containing GABA, theanine, melatonin, arginine and glutamine in the amounts listed below. Its components have separate preclinical literatures: GABA as the main inhibitory neurotransmitter of the cortex, theanine (gamma-glutamylethylamide) as a tea-leaf amino acid studied by rat brain microdialysis, melatonin as the pineal methoxyindole that tracks the light/dark cycle, and arginine and glutamine as precursors in nitric oxide and nitrogen metabolism. The references below cover the individual components, not this combination. Research Market supplies Sleep Mix with lot documentation on its certificate of analysis. It is sold strictly for laboratory research use.

Composition

  • GABA 100MG
  • THEANINE 50MG
  • MELATONIN 1MG
  • ARGININE 100MG
  • GLUTAMINE 25MG

Components

  • GABA (gamma-aminobutyric acid): CAS 56-12-2, C4H9NO2, 103.12 g/mol, PubChem CID 119
  • Theanine: CAS 3081-61-6, C7H14N2O3, 174.2 g/mol, PubChem CID 439378 (PubChem record for L-theanine; the isomer is not stated on the label)
  • Melatonin: CAS 73-31-4, C13H16N2O2, 232.28 g/mol, PubChem CID 896
  • Arginine: CAS 74-79-3, C6H14N4O2, 174.2 g/mol, PubChem CID 6322 (PubChem record for L-arginine; the isomer is not stated on the label)
  • Glutamine: CAS 56-85-9, C5H10N2O3, 146.14 g/mol, PubChem CID 5961 (PubChem record for L-glutamine; the isomer is not stated on the label)
  • Form: Fill amounts as listed above; physical form is stated on the vial label and batch certificate of analysis

Research applications

  • GABA: Reviewed as the main inhibitory neurotransmitter in the human cortex, including the contradictory evidence on whether orally supplied GABA crosses the blood-brain barrier [1]
  • Theanine and striatal neurotransmission: In conscious rats, theanine injected into the brain striatum raised glycine in microdialysis perfusate, and the glycine receptor antagonist strychnine reduced its effect on dopamine [2]
  • Melatonin physiology: Reviewed as a methoxyindole secreted mainly by the pineal gland at night, with a rhythm generated by the suprachiasmatic nuclei and entrained to the light/dark cycle [3]
  • Arginine metabolism: Reviewed as a precursor for nitric oxide, urea, polyamines, proline, glutamate, creatine and agmatine, with attention to the arginase and nitric oxide synthase isoenzymes [4]
  • Glutamine and immune cells: Reviewed from in-vitro and in-vivo studies as a nutrient for lymphocyte proliferation, cytokine production and macrophage activity, with circulating supply controlled by gut, liver and skeletal muscle [5]
