Compound monograph — Bronchogen

Bronchogen is the synthetic tetrapeptide Ala-Glu-Asp-Leu (AEDL), a respiratory-focused member of the Khavinson peptide series. The main in-vivo work is a rat model of chronic obstructive lung pathology induced by nitrogen dioxide, where bronchial epithelium structure and lavage fluid markers were measured. Biophysical papers on the Bronchogen peptide examine its binding to DNA and its effect on DNA melting temperature. Research Market supplies Bronchogen as a lyophilized research reagent, with purity and identity data for each lot reported on its certificate of analysis. It is sold strictly for laboratory research use.

Chemical structure and identification

  • PubChem CID: 11690869
  • Amino acid sequence: Ala-Glu-Asp-Leu (AEDL)
  • Form: Lyophilized powder
  • Analytical methods: RP-HPLC for purity and mass spectrometry for identity, reported on the batch certificate of analysis

Research applications

  • Obstructive lung model: In rats exposed to NO2 for 60 days, a month of bronchogen was reported to eliminate goblet cell hyperplasia, squamous metaplasia and loss of ciliated cells in bronchial epithelium and raised secretory IgA [1]
  • Lavage fluid markers: In the same NO2 rat model, bronchogen lowered neutrophilic inflammation in bronchoalveolar lavage and increased secretory IgA and surfactant protein B [2]
  • Bronchial epithelial differentiation: In human embryonic bronchial epithelial cell cultures, bronchogen stimulated expression of the differentiation factors CXCL12 and Hoxa3, more so in aged cultures [3]
  • DNA sequence preference: Fluorescence quenching assays indicated that bronchogen binds preferentially to deoxyribooligonucleotides with CNG sequences, in particular CTG [4]
  • DNA thermostability: Differential scanning microcalorimetry showed bronchogen raising the melting temperature of calf thymus and mouse liver DNA by 3.1 °C over a narrow peptide/DNA ratio range [5]
